Although Eci1p has a modified peroxisomal targeting sequence type 1 (PTS1, HisArgLeuCOOH) in its carboxyl terminus, its subcellular localization was confirmed. ECI1 was fused to the 3’ end of the gene encoding the green fluorescent protein (GFP) in a plasmid, and the linearized plasmid was integrated into the trp1 locus of wild-type yeast cells (strain BJ1991) and cells lacking peroxisomes (BJ1991pex6Δ) (Voorn-Brouwer et al. 1993). A fluorescence microscopy method was used to detect the localization of GFPs in yeast cells, as described earlier (Görner et al. 1998). The experiments were made in the Institute of Biochemistry and Molecular Cell Biology, University of Vienna, Austria.
